Determination of citalopram by HPLC in human plasma

Abstract:Objective To establish an HPLC-MS/MS method for determination of the content of citalopram in human plasma. Methods The chromatographic column was Agilent-ZORBAX-C8 (4.6×150 mm,5 μm), from Agilent Company, The mobile phase was methyl cyanides-0.1%formic acid(60∶〖KG-*2〗40). The flow rate was 0.5 ml/min and m/z 256.2167.1. The column temperature was 20 ℃. Electrospray ionization(ESI) source was applied and multi-reaction monitoring(MRM) mode with the transitions of m/z 325.1262.1(for citalopram) and m/z 256.2167.1 (for diphenhydramine internal standard)was used. Results The assay linearity of citalopram was confirmed over the range 0.5-100 ng/ml ( r =0.9979). The intra-and inter-day precision (RSD) was less than 10%. Conclusions The method is proved to be quick,specific,sensitive and accurate,which can be used for the determination of citalopram in plasma.
