Determination of three flavonoids in Apocynum venetum by high-performance capillary electrophoresis

Abstract:Objective To establish a high-performance capillary electrophoretic (HPCE) method for simultaneous determination of contents of rutin, isoquercitrin,and quercetin in Apocynum venetum. Methods A bare fused-silica capillary (58.5 cm×75 μm, effective length of 48.5 cm)was used. The buffer was composed of 50 mmol/L Na2B4O7 (pH 9.60). The running voltage was set at 20 kV with polarity from anode (inlet) to cathode (outlet). The separation time was 20 min and temperature was set at 25 ℃. Online detection was performed at 350 nm (PDA). Results The three types of flavonoids were well separated in 25 minutes and linear relationship in the peak area was good. The linear range was 4.4-44, 2.8-28.2, and 3.8-38 μg/ml, respectively. The correlation coefficient was 0.9968, 0.9995, and 0.9991, respectively. The average recovery was 97.3%, 98.4%, and 98.3%, respectively. Conclusions This method is simple, rapid, accurate, and reliable, which can be used for content determination of flavonoids in Apocynum leaves.
