Abstract:Objective To investigate the expression of interleukin-23(IL-23) and interleukin-17(IL-17) axis in the nasal polyp patients and explore the role of IL-23/IL-17 axis in the nasal polyp. Method Forty-five nasal polyp(NP) patients’s NP tissue and 30 simple deviation of nasal septum patients’ normal inferior turbinate tissue were collected. mRNA and protein expression of IL-23 and IL-17 were detected by immunohistochemistry and RT-PCR. SPSS 17.0 software package was used for statistical analysis. test and Spearman coefficient were used to analyse and compare the expression level and the correlation. Results RT-PCR demomstrated that the average expression level of IL-23p19mRNA and IL-17mRNA in NP tissues [(0.676±0.077),(0.681±0.044)] had significant differences (P<0.01) from those in normal inferior turbinate tissues [(0.212±0.015), (0.220±0.044)]. Immunohistochemical staining demomstrated that there were much more IL-23 and IL-17 positive cells in NP patients than in the normal controls. The average expression levels of IL-23 and IL-17 protein in NP tissues [(0.321±0.023), (0.315±0.023)] were significantly different (P<0.01) from those in normal inferior turbinate tissues [(0.199±0.011), (0.211±0.029)]. IL-17mRNA positively correlated with IL-23p19mRNA(r=0.723,P<0.01),and IL-17 protein positively correlated with IL-23 protein(r=0.991,P<0.01). Conclusion IL-23 and IL-17 may involved in the chronic inflammation of NP.IL-23 may be play a role through adjusting the level of IL-17.
