Abstract:Objective To investigate the expression and significance of SOX2 and PDCD4 in the tissues of pituitary adenoma.Methods A total of 120 patients with pituitary adenomas who were admitted to Chongqing Municipal Corps Hospital of Chinese People's Armed Police Force from June 2018 to March 2022 were selected, including 52 invasive pituitary adenomas and 68 noninvasive pituitary adenomas. The expression of SOX2 and PDCD4 was detected by immunohistochemistry; 10 cases of invasive pituitary adenoma tissues and 10 non-invasive ones were randomly selected, and the protein expression level was detected by western-blot. The value of SOX2 and PDCD4 in differentiating invasive pituitary adenoma and non-invasive pituitary adenoma was analyzed by ROC curve, and the area under the curve (AUC) was calculated. The factors affecting the invasiveness were analyzed by binary logistic regression model.Results The high expression rate and protein expression level of SOX2 in invasive pituitary adenoma tissues were 65. 38% and (1.32±0.13) respectively, which were higher than those in non-invasive pituitary adenoma tissues [27.94% and (0.38±0.08), P<0.05]. The high expression rate and protein expression level of PDCD4 in invasive pituitary adenoma tissues were 32. 69% and (0.47±0.10) respectively, which were lower than those in non-invasive pituitary adenoma tissues [51.47% and (1.09±0.11), P<0.05]. Tumor diameter, Ki-67 proliferation index, high expression of SOX2 and low expression of PDCD4 were the independent influencing factors for pituitary adenoma invasiveness (P<0.05). The AUC of SOX2 was 0.854 and PDCD4 was 0.783 in the differentiation between invasive and non-invasive pituitary adenomas, and the combination of SOX2 and SOX2 increased the AUC to 0.899.Conclusions The expression level of SOX2 is higher in the tissues of invasive pituitary adenoma, while the expression level of PDCD4 is lower, both of which have certain value in predicting the invasiveness of pituitary adenoma.
