Determination of monosaccharide composition in polysaccharides of Coreopsis basalis by gas chromatography |

Abstract Objective To establish a gas chromatographic method for determining the monosaccharide compositions and contents in polysaccharides of Coreopsis basalis. Methods Polysaccharides of Coreopsis basalis were derivatized after hydrolysis into monosaccharides with 4 mol/l TFA. Monosaccharide derivatives were separated on Shimadzu DM-5 column (30 m×0.25 mm,0.25 μm). The six monosaccharides were simultaneously separated under optimized conditions within 45 minutes. The calibration plots were linear over a range from 0.6 to 10 mg/ml. The recovery was 101.5%-103.4%. Results Polysaccharides of Coreopsis basalis consisted of Rha, Ara, Xyl, Man, Glu, and Gal with a molar ratio of 0.42∶2.51∶0.48∶0.83∶6.38∶2.07. Conclusions This method is simple, sensitive, accurate and reliable. It can be applied to quality control and determination of monosaccharide composition of Coreopsis basalis.
