Abstract Objective To study the efficacy and safety of donepezil hydrochloride combined with buflomedil in treatment of vascular dementia. Methods Fifty-four patients with vascular dementia were randomly divided into study group (n=27) and control group (n=27). In addition to the same routine treatment, patients in the study group were treated with donepezil hydrochloride and buflomedil, patients in the control group were treated with donepezil hydrochloride alone. Before and after treatment for 4 weeks, 12 weeks, 24 weeks, Mini-Mental State Examination (MMSE) and Activity of Daily Living (ADL) scales were used to assess the efficacy. Results After treatment for 12 and 24 weeks, the scores of MMSE and ADL were significantly improved in the study group and control group, compared with those before treatment(P<0.05). The scores and cure rates were significantly better in the study group than those in the control group(P<0.05). During the course of treatment, no obvious adverse reactions appeared. Conclusions Donepezil hydrochloride combined with buflomedil in treatment of vascular dementia is more effective than donepezil hydrochloride alone.
