Doxofylline inhibites U937 cells proliferation and induces apoptosis in vitro |

Abstract Objective To study the effects of the inhibitors of phosphodiesterase for acute myeloid leukemia in vitro culture U937 cell proliferation and apoptosis ,and provide basis for the clinical adjuvant treatment of leukemia. Methods Add Doxofylline of different experimental concentrations was added respectively to a certain concentration of U937 cells in ogarithmic phase. Cell proliferation rate was detected by WST-1 method and the apoptosis by TUNEL method. Results Different concentrations of doxofylline acted on U937 cells respectively. The degree of inhibition of cell proliferative activity gradually increased with prolonged action time and the increase of drug concentration(P<0.01), the most obvious among them was in 60 mg/L group. Different concentrations of doxofylline promoted U937 cell apoptosis and apoptosis with significant concentration dependence and time dependence(P<0.01). Conclusions Confirmed by the experiment, doxo fylline can inhibit leukemia U937 cell line proliferation and induce its apoptosis,and has concentration dependence and time dependence.
