The status quo and molecular evolution of occult infection in intrafamilial patients with hepatitis B virus infection in Wuwei |

Abstract Objective To study the status quo and molecular evolution of occult infection in intrafamilial patients with hepatitis B virus infection in the city of Wuwei.Methods All the people who were found to be infected with HBsAg during a physical examination in 2009 in a town in Wuwei were questioned for their HBV family history. After they signed informed consent, their blood was drawn and tested for hepatitis B surface antigen (HBsAg), antibody to HBsAg (anti-HBs), hepatitis B e-antigen (HBeAg), antibody to HBe (anti-HBe) and antibody to hepatitis B core antigen (anti-HBc) using commercially available enzyme-linked immunosorbent assay (ELISA). DNA was extracted using magnetic Beads extraction kit from all the samples, and amplification was performed by nested PCR. Sequences obtained were analyzed by mega 6.0.Results Of the 80 serum samples, the positive rate of HBsAg, Anti-HBc, and HBV DNA was 43.8%, 78.8% and 37.5%, respectively. The rate of infection with intrafamilial occult hepatitis B was 26.7%(12/45).Conclusions The prevalence of intrafamilial occult hepatitis B infection is significantly higher than in the general population. The main genotype is genotype C and genotype D in particular.
