Elevated serum and urinary miRNA- 150 and miRNA-30a-5P levels in children with idiopathic nephrotic syndrome |

Abstract Objective To analyze the difference of expressions of miRNA-150 and miRNA-30a-5P in serum and urine of children with idiopathic nephrotic syndrome (INS)and discuss their clinical value as a non-invasive auxiliary diagnostic marker of INS.Methods Urine samples were collected from 93 INS children (the patient group) who were diagnosed but waited to be treated in our hospital between January 2017 and December 2019 and from another 89 sex- and age-matched healthy children (the control group). The levels of serum and urinary miRNA-150 and miRNA-30a-5P were determined by quantitative real-time polymerase chain reaction (qRT-PCR) . The clinical value of serum and urinary miRNA-150 and miRNA-30a-5P in the diagnosis of children’s INS was assessed by the ROC curve.Results The levels of serum and urinary miRNA-30a-5p and serum miRNA-150 were slightly higher in the patient group than in healthy controls (P<0.05) , but the levels of urinary miRNA-150 were not. The levels of serum and urinary miRNA-30a-5P and serum miRNA-150 in the clinical remission period decreased significantly in forty-six of these cases(P<0.05) .The area under the ROC curve (AUCROC) of serum miRNA-150 and miRNA-30a-5P was 0.852 and 0.869 respectively for the diagnosis of INS children, compared to 0.825 and 0.833 for urinary miRNA-150 and miRNA-30a-5P.Conclusions The serum and urinary levels of miRNA-150 and miRNA-30a-5P in children with idiopathic nephrotic syndrome increase obviously, which may serve as new biopsy molecular markers for the auxiliary diagnosis of INS.
