Effects of calcium hydroxide in combination with triple antibiotiques on Enterococcus faecalis virulence factors |

Abstract Objective To determine the effects of the combination of triple antibiotic paste (TAP) with calcium hydroxide paste (CH) on expressions of virulence genes(gel E, cyl A and esp) of E.faecalis biofilms.Methods A model of E.faecalis biofilms in the starvation phase was established in vitro. Samples were randomly divided into four groups: normal saline (NS)control group, calcium hydroxide paste (CH)group, triple antibiotic paste (TAP)group, and combination(TAP+CH)group. The effects of the three medicines on expressions of virulence genes(gel E, cyl A and esp) in E.faecalis were evaluated by quantitative polymerase chain reaction.Results After treatment, the three experimental medications all inhibited the expressions of virulence genes (gel E, cyl A and esp) more significantly than in the control group (gel E:22.74±1.63,13.23±1.26,10.68±1.03 vs. 38.65±3.00, P<0.05; cyl A:8.60±0.68, 7.05±0.55, 6.34±0.30 vs. 10.91±1.00, P<0.05; esp:10.65±1.29, 7.05±0.55, 6.01±0.44 vs. 13.97±2.42, P<0.05). TAP+CH and TAP both inhibited the expressions of virulence genes more significantly than CH did. There was no significant difference between TAP+CH and TAP in the expressions of these genes.Conclusions TAP+CH and TAP play an important role in inhibiting biofilm formation.
