Effects of Xuejie and Qingdai alone and in combination on skin wound healing in diabetic rats |

Abstract Objective To investigate the effect and mechanism of Xuejie and Qingdai and in combination on skin wound healing in diabetes rats. Methods The diabetic rat model was prepared and randomly divided into normal control group, model control group, Xuejie group, Qingdai group and Xuejie-Qingdai compatibility group, with 6 rats in each group. After 14 days of administration, the cervical vertebrae were removed for execution, and the healing rate of the wounds was calculated, and H-E staining was used to observe the pathology of the skin; RT-PCR was used to detect the expression of the factors related to the TGF-β1/Smads signaling pathway. Results Compared with the model control group, the Qingdai group had a lower skin wound healing rate, while the Xuejie group and combination group showed a significant improvement in healing rate (64.19% and 61.16%). The pathological observation of HE staining suggested that the epidermal integrity of the skin tissues in the Xuejie group and the combination group were better than that the Qingdai group. The newly formed epidermal layer was thicker, the cell arrangement was irregular, and there were more newly formed capillaries and a small amount of inflammatory cell infiltration. Compared with the model control group, the expression of TGF-β1 mRNA increased in each intervention group (P<0.05); The expression of Smad2 and Smad3 mRNA in the Xuejie group (5.204±1.070, 1.201±0.052)was higher than that in the model control group; the expression level of Smad 7mRNA was upregulated by using only Xuejie or Qingdai(5.125±1.918, 6.896±3.628), while its expression was downregulated by their combination[(5.125±1.918), (6.896±3.628)]; the expression of ERK1/2 in the Xuejie and Qingdai monotherapy groups were not significantly different from that in the model group, but there was statistically significant difference between the combination group(3.664±0.357) and the model group (P<0.05). The expression of JNK and p-38MAPK mRNA in the Qingdai group was higher than the model group (P<0.05). Conclusions Xuejie and Qingdai can promote skin wound healing in diabetes rats, and the mechanism may be related to regulating the expression of TGF-β 1/Smads signal pathway related factors.
