HPV E6 promotes invasion and metastasis of cervical cancer cells by regulating expression of miRNA-145

Abstract:Objective To study the molecular mechanisms of cervical cancer HPV infection regulating host cells miRNAs’ expression and its significance in invasion and metastasis of cervical cancer. Methods Northern blotting was used to detect the impact of over-expression of p53 on the expression of miR-145 in cervical carcinoma cells, compared with the impact of lowering HPV E6 on miR-145 expression. Western blotting was used to detect the impact of over-expression of the E6 on endogenous and exogenous p53 expression and influence of knockdown of HPV E6 cervical cancer cells on exogenous p53 expression. Scratch test was used to detect the different expressions of miR-145 in HeLa cells and the female parent HeLa cells in cell migration ability. Trans-well test was used to observe differences in the over-expression of miR-145 in HeLa cells and the female parent HeLa cell invasion. Results In cervical cancer cells, over-expression of p53 significantly increases expression of miR-145; over-expression of HPV E6 reduces the expression of p53 in cervical cancer cells; knockdown of HPV E6 expression in cervical cancer cells enhances the expression of exogenous p53 and the expression of miR-145 induced by hydrogen peroxide; over-expression of miR-145 inhibits cell migration and invasion of cervical cancer cells. Conclusions HPV E6 promotes the invasion and metastasis of cervical cancer cells, by inhibiting P53 and lowered the expression of miR-145.
