Abstract:Objective To investigate the effect of polyphyllin on the proliferation of osteoblast-like cells MC3T3-E1 and possible mechanisms. Methods The osteoblast-like cells MC3T3-E1, were administered with different concentrations of polyphyllin (3, 10, 30, and 100 μg/ml). Then, MTT colorimetric test, EdU kit, and alkaline phosphatase (ALP) activity detection kit were used to observe the proliferation of MC3T3-E1. Western blot analysis was employed to observe the expression of β-catenin and bone morphogenetic protein -2 (BMP-2) in MC3T3-E1 cells treated with 30, and 100 μg/ml of polyphyllin. Results Polyphyllin could effectively promote proliferation of MC3T3-E1 cells and upregulate the expression of β-catenin and BMP-2 expression. Conclusions Polyphyllin may promote MC3T3-E1 cell proliferation and differentiation via regulating β-catenin and BMP-2 signaling pathway.
