Abstract:Objective To study the effect of arsenic trioxide on inducing apoptosis of osteosarcoma U2OS cells in vitro. Methods Osteosarcoma U2OS cells were treated with arsenic trioxide at different concentrations(0,4 μmol/L,8 μmol/L) for 24 h, respectively, then were observed under microscope,by MTT assay and Hochst33342 staining method. U2OS cells were treated with arsenic trioxide at 8 μmol/L and then detected by flow cytometry at 8 h, 16 h and 24 h. Results AS2O3 could inhibit U2OS cells growth, the U2OS cells proliferation rate was (66.32±5.15)% after treatment with arsenic trioxide at 4umol/L concentration, and (26.73±9.36)% at 8 μmol/L concentration. The U2OS cells apoptosis rate was 21.16%,36.15%,47.13%respectively at 8 h, 16 h, 24 h after treatment with AS2O3. The effect of arsenic trioxide on inhibiting U2OS cells growth is dependent on time and concentration. Conclusions AS2O3 can induce apoptosis of osteosarcoma U2OS cells and inhibit the cells growth.
