Effect of extracts of Picorhiza scrophlariiflora Pennell on renal tubular epithelial cells with hypoxia reoxygenation |

Abstract Objective To investigate the mechanism of oxidative stress of renal tubular epithelial cells with hypoxia reoxygenation (H/R) and to evaluate the effects of extracts of Picorhiza scrophlariiflora Pennell on them. Methods The renal epithelial cells were reoxygenated after hypoxia for 0.5h,1h or 2h to set up H/R cell model.The renal epithelial cells were divided into the following groups: normal control grounp,H/R group, 10 μg/ml extracts of Picorhiza scrophlariiflora Pennell+H/R group, and 100 μg/ml extracts of Picorhiza scrophlariiflora Pennell+H/R group, 1000 μg/ml extracts of Picorhiza scrophlariiflora Pennell+H/R group. The flow cytometry was used to detect the oxidative and the apoptosis of renal tubular epithelial cells Result The oxidative stress in renal tubular epithelial cells was low. The amounts of apoptosis and necrosis were small.The oxidative stress in renal tubular epithelial cells was induced and increasd after H/R, compared with that in normal HK2 cells(P<0.05). The amounts of apoptosis and necrosis were increasd after H/R, compared with that in normal HK2 cells(P<0.05).Oxidative stress in renal tubular epithelial cells after H/R was inhibited by extracts of Picorhiza scrophlariiflora Pennell in a dose-dependent manner(P<0.05). The apoptosis and necrosis of renal epithelial cells after H/R were inhibited by extracts of Picorhiza scrophlariiflora Pennell in a dose-dependent manner(P<0.05). Conclusion Extracts of Picorhiza scrophlariiflora Pennell protect renal epithelial cells against apoptosis and necrosis by inhibiting oxidation after H/R injury.
