A Study on XAV939’s function in inhibiting ovarian cancer cells migration and invasion |

Abstract Objective To investigate the effects of XAV939 on invasion and metastasis of ovarian cancer cells as well as the inhibition of the Wnt / β-catenin signaling pathway. Methods The MTT assay was used to detect the inhibition effect of XAV939 on the viability of ovarian cancer, the inhibition of cell metastasis was detected by scratch test and transwell cell migration assay, the expression level of β-catenin,wnt1,TCF4 and MMP9 protein by Western blotting.Results The MTT assay showed that the inhibition rate of ovarian cancer increased significantly by XAV939 for 48 h,especially the concentration of 8 μmol/L(31%±1%,P=0.034),16 μmol/L (51%±4%,P=0.028). Scratch test and transwell cell migration assay showed the inhibition of invasion and metastasis after XAV939 treatment. In addition, the results of Western blotting showed that XAV939 reduced the expression of wnt1,β-catenin,TCF4,and MMP9 protein. Conclusion XAV939 can inhibit the invasion and migration of ovarian cancer cells through Wnt/β-catenin pathway.
