Effect of resistant dextrin on gut microbiota in rats with chronic unpredicted mild stress |

Abstract Objective To explore the effect of resistant dextrin on gut microbiota in rats with chronic unpredicted mild stress.Methods Twenty-four male SD rats were randomly divided into a control group, a model group and a resistant dextrin group. A depression-like rat model was established using chronic unpredictable mild stress (CUMS). After 3 weeks of stress modelling, sucrose preference test and open- field test were used to evaluate the depression-like behavior of rats. Stress sensitive rats were screened in the model group, and the same number of rats in the other two groups were randomly selected to detect the composition and structural changes of gut microbiota by 16S rRNA sequencing.Results Compared with the control group, relative abundance of Firmlcutes [(0.92251±0.02838) vs. (0.82572±0.00761)], Akkermansia[(0.01327±0.00076) vs. (0.00000±0.00000)]and Lactococcus [(0.00029±0.00010) vs. (0.00000±0.00000)] in the model group was significantly reduced (P<0.05); relative abundance level of Bacteroidetes (0.04671±0.00263) vs. (0.12341±0.03576), Faecalibacterium[(0.00011±0.00004)vs.(0.00057±0.00005)]and Ruminococcaceae_UCG_014 [(0.01003±0.00384)vs.(0.10976±0.04364)] was significantly increased (P<0.05). Compared with the model group, resistant dextrin could significantly callback the microflora with changed abundance (P<0.05).Conclusions Resistant dextrin can regulate the gut microbiota of CUMS rats and restore intestinal dynamic balance by improving the relative abundance of probiotics and reducing the relative abundance of pathogenic bacteria, which provides experimental basis for the treatment of depression.
