Ets-1在不同Banff分级慢性肾脏病肾组织中的表达

摘要目的探讨不同Banff分级慢性肾脏病患者肾脏中Ets-1(V-ets erythroblastosis virus E26 oncogene homolog 1)表达的差异。方法选取在本院进行肾穿刺活检的患者60例,根据患者肾脏纤维化程度,分为Banff分级正常、轻度、中度及重度4组,每组各15例。对4组患者的性别、年龄、生化指标及病理特征进行分析,并通过免疫组化方法检测肾组织中Ets-1的表达。结果 Banff分级轻度组的BUN、UA、iPTH、尿β₂-MG较正常组增高;Banff分级中度组的BUN、SCr、UA、iPTH及尿β₂-MG较正常组增高;Banff重度组的MABP、BUN、SCr、UA、血磷、iPTH、尿β₂-MG均较正常组明显升高,而Hb降低。Banff轻度、中度及重度组患者球性硬化、小管萎缩、间质炎性细胞浸润及间质纤维化比例均显著高于正常组。免疫组化显示Banff分级正常组、轻度组、中度组及重度组Ets-1的 A值分别为0.10±0.01、0.15±0.08、0.32±0.07、0.54±0.12,中度组、重度组较正常组升高。结论肾间质纤维化患者Ets-1表达升高且随Banff分级程度加重而增高。

Abstract：Objective To investigate the expression of transcription factor Ets-1 in chronic renal fibrosis.Methods A total of 60 patients underwent renal biopsy from 1 May 2013 to 31 May 2015 at the Department of Nephrology in The Third Affiliated Hospital of Suzhow University were enrolled in the study and divided into four groups according to Banff scheme. Clinical and pathological characteristics of patients were investigated.The expression of transcription factor Ets-1 was assessed by immunohistochemical stainining.Results Compared with normal group,serum BUN, UA, iPTH and urineβ₂-MG were significantly elevated in Banff Ⅰgroup. Serum BUN、SCr、UA、iPTH and urineβ₂-MG were significantly elevated in Banff Ⅱgroup. MABP、BUN、SCr、UA、P、iPTH and urineβ₂-MG were significantly elevated in Banff Ⅲgroup while Hb decreased. Percentages of global glomerulosclerosis,tubular atrophy, inflammatory cell infiltration and interstitial fibrosis significantly increased in Banff Ⅰ,Ⅱ,Ⅲgroups than in normal group. Immunohistochemical analysis showed that absorbance values of A were 0.05±0.01, 0.15±0.08, 0.32±0.07, and 0.54±0.12, respectively. Ets-1 were up-regulated in Banff Ⅱand Ⅲ groups.Conclusion Ets-1 is up-regulated in renal fibrosis and increases with Banff schems.
