A study on optimize formulation and technique of asiaticoside non-ionic surfactant vesicles

Abstract:Objective To optimize the formulation and preparation technique of asiaticoside non-ionic surfactant vesicles.Methods The thin film evaporation method was used to prepare the asiaticoside niosomes, then taking encapsulation efficiency and drug loading rate as an indicator, a single factor experiment was conducted including prescription and technique factors such as the type and amount of surfactant, dosage of the drug, drug-lipid ratio, incubation time, incubation temperature, speed of the rotary evaporator and so on. The optimized asiaticoside niosomes formulation and preparation process was acquired by orthogonal method and was further verified.Results The results of single factor study showed that the three factors which had greater impact on encapsulation efficiency and drug loading rate were surfactants and the cholesterol ratio (m/m), incubation temperature and incubation time. The optimized asiaticoside niosomes formulation is as follows: asiaticoside dosage is 36 mg with the ratio of lipids as 4∶1, incubation temperature is 30 ℃, incubation time is 30 min.The encapsulation efficiency and drug loading rate of the asiaticoside noisome which was prepared according to the optimized formulation was 89.56% and 25.26% and with a microcosmic morphology of typical smooth vesicles.Conclusions The asiaticoside niosomes prepared by thin film evaporation method shows a well-rounded appearance with good dispersity and rather high encapsulation efficiency and drug loading rate.
