Simultaneous determination of illegal dyes of New fuchsin,Auramine O and Sudan Ⅳ in Carthami Flos by LC-MS/MS

Abstract:Objective To develop an LC-MS/MS method for simultaneous determination of illegal dyes of New fuchsin, Auramine O and Sudan Ⅳin Carthami Flos.Methods Using Agilent EC-C18(2.1 mm×50 mm,1.8 μm)column, the mobile phase consisted of 20 mmol/L ammonium formate+0.1% methane acid-acetonitrile using gradient elution. The column temperature was 40 ℃, ionization mode was ESI, and detection mode was MRM. The ion pair selected was: ①New fuchsin m/z 330.3→223.2,m/z 330.3→300.2; ②Auramine O m/z 268.2→147.2,m/z 268.2→252.2; ③Sudan Ⅳ m/z 381.2→224.1,m/z 381.2→91.1.Results The linear range of New fuchsin, Auramine O and Sudan Ⅳ was 0.1086-271.50 ng/ml, 0.1392-348.00 ng/ml and 0.1352-338.00 ng/ml, respectively, the recoveries were 97.45%, 96.44% and 97.58%, respectively, and RSD was 0.70%,1.12% and 0.89%, respectively. RSD of precision, stability and repeatability was less than 3%.Conclusions This method of extraction is simple to use, accurate for determination and highly sensitive. This method can be applied to the determination of illegal dyes of New fuchsin, Auramine O and Sudan Ⅳ in Carthami Flos.
