Abstract:Objective To improve the accuracy of detection of peripheral blood lymphocyte subsets by flow cytometry. Methods Problems that were often encountered by our laboratory in the process of detecting peripheral blood lymphocyte subsets with the Mindray's BriCyte E6 flow cytometer in 2018 were summarized and analyzed, and corresponding solutions were proposed. Results The common problems encountered in the detection of peripheral blood lymphocyte subsets by flow cytometry included unclear clusters of scatter plots of the CD45-SSC gate, unclear classification clusters of scatter plots of the cross gate, inability to classify lymphocyte subsets, and false reduction of absolute lymphocyte subsets counting. The above problems could be solved by washing the blood samples with saline before sample preparation or by cleaning the samples with hemolysin before detection. Conclusions The problems encountered in the detection of peripheral blood lymphocyte subsets by Mindray BriCyte E6 flow cytometry can be solved by washing the blood samples with saline or cleaning the samples with hemolysin.
