Effect of specimen disposal on detection of glycosylated hemoglobin by OLIMPO specific protein analyzer |

Abstract Objective To explore the effect of sample hemolysis time and preservation conditions on the detection of glycosylated hemoglobin (HbA1c) by OLIMPO.Methods 1.Anticoagulant samples of fresh whole blood were taken from ten patients to be preprocessed (hemolyzed). Sixty samples of 150 μl each were collected and equally divided into 6 groups at room temperature. Group A was immediately detected, while groups B, C, D, E and F were detected 1, 2, 3, 4 and 5 hours later, respectively. 2. After 5 samples of whole blood were mixed properly, they were divided into 4 groups with 5 samples in each. Group A1 was stored at room temperature (18-22 ℃), group B1 was refrigerated (2-8 ℃), group C1 cryopreserved (-18--20 ℃) , and group D1 was cryopreserved at a low temperature (below -80 ℃).The results of D1 were taken as the standard to detect the quality of each group with OLIMPO. The HbA1c results of each specimen were regarded as acceptable if they were within the range of ±3 compared with the results of D1.Results 1.HbA1c results at different time points after hemolysis were (7.98±1.61)% in group A, (7.97±1.61)% in group B, (7.95±1.60)% in group C, (7.94±1.61)% in group D, (7.77±1.61)% in group E and (7.46±1.57)% in group F. There was statistically significant difference between group A and group F (t=1.57,P<0.05). 2. In order to keep HbA1c results stable during OLIMPO test, the storage time of samples was 2D at room temperature, 15 d for cold storage, 3D for cryopreservation and more than half a year for cryopreservation at a low temperature.Conclusions When HbA1c is detected with OLIMPO, the stability varies considerably under different processing conditions. The results are relatively accurate within 4 hours after sample hemolysis. If the whole blood sample specimen needs to be stored for a longer time (for scientific research or re examination), better preservation can be achieved at 2-8 ℃ and -80 ℃.
