Effect of tacrolimus and cyclosporine on glucose metabolism in rats |

Abstract Objective To compare the effect of tacrolimus and cyclosporine on glucose metabolism in rats. Methods SD rats were randomly divided into 3 groups. Rats in control group were administrated with distilled water 1 ml/(kg·d). Those in cyclosporine group were administrated with cyclosporine 25 mg/(kg·d) while those in tacrolimus goup were administrated with tacrolimus 2 mg/(kg·d). The medicine was administrated through the stomach tube. The experiment lasted 5 months. Fasting blood glucose levels and insulin levels were tested at the end of the experiment. The insulin resistance index and insulin sensitivity index were calculated. Results After 5 months of treatment, fasting blood glucose levels and the insulin resistance index in cyclosporine group and tacrolimus group were higher than those in control group(P<0.05, individually). The insulin sensitivity index in cyclosporine group and tacrolimus group was lower than that in control group(P<0.05, individually). The rats in tacrolimus group showed higher blood glucose levels, a higher insulin resistance index and lower insulin sensitivity index than those in cyclosporine group(P<0.05, individually). Conclusion The therapeutic dose of tacrolimus or cyclosporine has effect on blood glucose metabolism. Tacrolimus has better hyperglycemic effect than cyclosporine.
