Abstract:Objective To explore the effects of platelet-rich plasma (PRP) on the proliferation and differentiation of skin cells and its mechanisms of action. Methods Human dermal fibroblasts(HDFs)and human epidermal keratinocytes(HEKs) were cultured in vitro and subjected to varying concentrations of PRP at different concentrations. Cell counting kit-8(CCK-8), immunofluorescence staining and enzyme-linked immunosorbent assay(ELISA) techniques were utilized to evaluate cell proliferation, differentiation, and the expression of associated growth factors, and the effects of different concentrations of PRP on HDFs and HEKs were systematically evaluated. Results PRP significantly enhanced the proliferation rates of HDFs and HEKs,facilitated cell differentiation,and upregulated the expression of growth factors such as platelet derived growth factor(PDGF),epidermal growth factor(EGF),and fibroblast growth factor(FGF) , and the differences were statistically significant(P<0.05). Conclusions PRP can increase the expression level of growth factors in promoting the proliferation and differentiation of skin cells.
